Is this possible for someone to write a rootkit with Ctypes?
Not a rootkit on its own, but here's a process injector I wrote that allows you to inject shellcode into a running process in order to hide its execution.

To answer the question though, you cannot definitively prove this. If the keylogger installed itself as a rootkit, the OS may not even be aware the code is running. There are ways of completely hiding programs by rewriting the linked list the OS uses to maintain the process list. Or, it could be running inside another process using something like this proof of concept I wrote a while ago.

Code that attempts to read and interact with the memory-space of other programs can be used to do stuff like this.

Are there any cases for using C to interact with Python?
Here's a program I wrote to do process injection that interfaces with the Windows C API to control process's memory. If you need to do low-level OS manipulation, you either need to use something like ctypes or use an existing library that does the same.

ctypes allows you to interface with, for example, the Windows API to do things like process injection. At least that's the route I went to do that.

What do I need to learn to create a cheat/fake cheat software for games?
The basics of what you want though are accomplished by using the WinApi. Really every language is capable of interfacing with the API, so the language doesn't really matter. I've even interfaced with it in Python.
