As a stab in the gloom, I think it has to do with the Qt library. On the Calibre change log it mentions that Calibre 6 now uses Qt 6, and Qt 5 plugins may not work. In the Github page for the DeDRM project, the file config.py is trying to load Qt 5. It also looks like DeDRM hasn't been updated since about the time Calibre 5 came out.
noDRM has picked up the project. There's an update a few days ago for Calibre 6 and Qt 6. Github repo is at https://github.com/noDRM/DeDRM_tools
