How to use Windows ssh-agent on a WSL2 distribution to avoid entering SSH key passphrases?
Are you talking about npiperelay? In the README you'll see you need to install socat in WSL2 to communicate with it. Sorry if I've misunderstood.

SSH from a container?
Then, for some special situations (e.g. running a suite a few tools that are useful for me and some other devs on my team), the quick solution is to simply just setup a Dockerfile based on alpine:latest and install the other tools/apps I need that require SSH key forwarding, like capistrano. I already have putty agent running on my host machine with ssh-pageant which connects my Cygwin instance to that. But anyway, on the container, I just setup basic sshd there, login with keys and enable agent forwarding and then run my tools there that then hop on to the remote server (using my forwarded agent).
What are some alternatives?
When comparing npiperelay and ssh-pageant you can also consider the following projects:
wsl2-ssh-pageant - bridge between windows pageant and wsl2
wsl-ssh-pageant - A Pageant -> TCP bridge for use with WSL, allowing for Pageant to be used as an ssh-ageant within the WSL environment.
wsl-ssh-agent - Helper to interface with Windows ssh-agent.exe service from Windows Subsystem for Linux (WSL)
OmniSSHAgent - Integrated ssh-agent for windows. (pageant compatible. openSSH ssh-agent etc ..)
wetty - Terminal in browser over http/https. (Ajaxterm/Anyterm alternative, but much better)
keepassxc - KeePassXC is a cross-platform community-driven port of the Windows application “Keepass Password Safe”.
